The research project "Design of supramolecular architectures based on metal derivatives of phthalocyanine-functionalized nanoparticles with potential for applications in medicine" focused on the development of new photosensitizers in injectable solution that would possess improved photodynamic properties. The main objective was the synthesis of zinc phthalocyanine with four groups and eight groups (COOH), respectively, with the formulas ZnPc(COOH)4 and ZnPc(COOH)8, respectively their conjugation with magnetic Fe3O4 nanoparticles.
The derivatives ZnPc(COOH)4 and ZnPc(COOH)8 were synthetized, which were then conjugated with chitosan and dextran and photosensitizers (FS) for photodynamic therapy ZnPc(COOH)4/Dx and ZnPc(COOH)8/Dx, ZnPc(COOH)4/Chs and ZnPc(COOH)8/Chs were obtained. Also, Fe3O4 nanoparticles were synthetized by the coprecipitation method, then, they were functionalized with chitosan, Fe3O4/Chs and conjugated with ZnPc(COOH)4 and ZnPc(COOH)8 derivatives.
The photophysical parameters of the ZnPc(COOH)8/Fe3O4/Chs FS were determined and compared with those of ZnPc(COOH)4/Fe3O4/Chs. It was found that the FS with the ZnPc(COOH)8 derivative exhibits a longer singlet lifetime than the FS with ZnPc(COOH)4, while the quantum yield of both fluorescence and phosphorescence is high for the FS ZnPc(COOH)4, and with increasing concentration of the ZnPc(COOH)4 derivative, the singlet lifetime increases from 3.76 ns to 3.81 ns, the fluorescence quantum yield changes from 12.81% to 15.86%.
The photophysical parameters of the ZnPc(COOH)8/Fe3O4/Chs supramolecular architectures were measured and compared with those of ZnPc(COOH)4/Fe3O4/Chs. Zinc octachloromethyl phthalocyanine (ClMe)8ZnPc was synthetized according to the chloromethylation reaction of ZnPc. Next, the zinc phthalocyanine octakis (methyl-isothiouronium chloride) (MeIt)8ZnPcCl8 and the zinc phthalocyanine derivative with eight thiol groups (HSMe)8ZnPc were obtained.
AgNPs were synthetized by the chemical reduction method using chitosan and tannic acid as reducing agents, and glucose, sodium citrate, ascorbic acid as stabilizers. These were conjugated with the zinc phthalocyanine derivative with eight thiol groups. |
